Motivation

Photoacoutic microscopy (PAM) is based on tissues absorption, using a supercontinuum source is a ultra-broad band (typically 500-2400nm) may be relevant for multispectral PAM and permits to image multiple absorbers within a sample and give the oxygen saturation of the imaged blood vessels for instance. For PAM the supercontinuum source need to be filtered within a relatively small bandwidth (<50nm). However current supercontinuum sources have too low power to be used for multispectral PAM.

I worked at NKT Photonics on building a supercontinuum source with sufficient energy (>50nJ) and bandwidth (<20nm) for multispectral PAM. This single source has already proved its usefulness for multispectral PAM (<2nJ, <15nm) on ex-vivo retinal pigment epithelium and blood samples, and in-vivo PAM on a mouse ear (120nJ, 250nm). This supercontinuum source has been used as well for OCT on in-vivo mouse ears.

Recent developments were focused on making the supercontinuum source robust with sufficient energy for in-vivo combined multispectral PAM and OCT. The dual modality system is currently in the process of being build at the University of Kent.

The source has more than 150 nJ per 10 nm bandwidth over a spectral range of 500 nm to 1600 nm. This performance is achieved using a carefully designed fiber taper with large-core input for improved power handling and small-core output that provides the desired spectral range of the supercontinuum source.

  • Abstract: We report the usefulness of a single all-fiber-based supercontinuum (SC) source for combined photoacoustic microscopy (PAM) and optical coherence tomography (OCT). The SC light is generated by a tapered photonic crystal fiber pumped by a nanosecond pulsed master oscillator power amplifier at 1064 nm. The spectrum is split into a shorter wavelength band (500– 800 nm) for single/multispectral PAM and a longer wavelength band (800– 900 nm) band for OCT. In vivo mouse ear imaging was achieved with an integrated dual-modality system. We further demonstrated its potential for spectroscopic photoacoustic imaging by doing multispectral measurements on retinal pigment epithelium and blood samples with 15-nm linewidth.

    • Abstract: We report on the use of a single supercontinuum (SC) source for multimodal imaging. The 2-octave bandwidth (475-2300 nm) makes the SC source suitable for optical coherence tomography (OCT) as well as for multispectral photoacoustic microscopy (MPAM). The IR band centered at 1310 nm is chosen for OCT to penetrate deeper into tissue with 8 mW average power on the sample. The visible band is used for MPAM. The source is equipped with the ability to select the central wavelength as well as the spectral bandwidth. An energy of more than 35 nJ within a less than 50 nm bandwidth is achieved on the sample for wavelengths longer than 500 nm. In the present paper, we demonstrate the capabilities of such a multimodality imaging instrument based on a single optical source. In-vitro mouse ear B-scans images of a mouse ear are presented.

An energy of 5 to 40 nJ is achieved on sample within a 50 nm bandwidth in the visible range in the photoacoustic channel. Also, a few mW power is also achieved on the sample in the optical coherence tomography channel.

    • Abstract: Supercontinuum sources are very suitable for many applications involving high resolution biophotonics imaging such as photoacoustic imaging. Using several wavelengths simultaneously, for example in the process of photoacoustic imaging, allows for more information to be acquired from a sample. However, at the moment the lack of pulse energy delivered by a supercontinuum source is an important limitation in improving system resolution and imaging depth. To generate a supercontinuum spectrum we pump a highly nonlinear fiber. Its small core size limits the input pulse energy and therefore the supercontinuum source output energy. Unfortunately we cannot use a larger core fiber since doing so will mitigate or fully prevent manifestation of the nonlinear effects needed to generate a broadband spectrum. The initial focus of this PhD project is to investigate the coupling of a supercontinuum pump source with relatively high peak power into a small core fiber without incurring damage to that fiber using, for example, tapered fiber.
